⚙️ Calibration

Calibration of testing equipments is important for optimal functioning and accurate parameter readings. It is highly recommended to calibrate the equipments and probes at least once a month and note it down on the husbandry sheet. The details for calibrating various equipments and probes used in the lab are as follows:

1. HANNA probe

Calibration instructions are documented here. Extra battery in case of low battery in the device can be found at the Kao Lab Room 5350. The HANNA pH and EC/TDS calibration solutions are kept in the Kao Lab refrigerator in the animal facility room 5550. When using these solutions ensure that you utilize the minimum amount of solution required and use appropriate measuring cylinders and glass ware to pour in the solutions from the bottles.

2. Oxygen probe

Instructions for calibrating the oxygen probe can be found here. In the manual it specifies to dip the probe in the MA9070 zero oxygen solution. This solution can be prepared by the user by mixing the zero oxygen solution component 1 and DI water. To ensure re-usability we recommend dissolving 1.2gm of component 1 in 50mL of DI water (can be collected from DI water tap installed in the lab). Again, make sure to use appropriate measuring cylinders and glass ware to pour in make the solutions.

💡 The probe should always be checked for any imperfections on the membrane (bottom part of the probe). The membrane is quite delicate and should always be smooth and without air bubbles underneath it using the small screwdriver provided with the equipment. Inform the lab personnel in case we are running low with membranes or solutions.
